There are no special requirements to rent a car in Czech, as long as you have a valid drivers license you should be fine. Depending on your arrival airport you may be renting a car in Poland or Austria instead, please note that in Austria and Poland an international drivers permit (obtained in the US from AAA) is necessary if stopped by the police. The rental car company won't require it, but if you get pulled over technically you are required to have one.
The only option is by rental car. Please use google maps to determine the best route depending on what airport you arrive at. Note that many highways have tolls, your rental car company will typical handle payment and send you a bill after you return the car.
Thankfully the EU has pushed back the implementation ETIAS, which would of required American's to have a visa for EU travel to 2025. As such there are no special requirements for US travelers to enter the EU in 2024. However make sure your passport is valid and will not expire for at least 3 months after leaving the EU. If it expires within 3 months of your flight you may have issues entering the EU.
