Flight
If you're planning on coming straight to the Peninsula, there's the Peninsula Express (MEL) SkyBus that goes from the airport and has multiple stops on the Peninsula. If you're planning on staying in Melbourne first, there are 'Turn Up & Go' SkyBus services into the city 7 days per week on the Melbourne City Express (MEL), 4AM - 1AM.
Train
If you've decided to explore Melbourne City first, you can get the train from Flinders Street Station or Southern Cross Station to Frankston Station on the Peninsula. From there, you can either get a local bus or taxi (available outside Frankston Station) to your accommodation.

For the wedding weekend we suggest looking for accommodation in the following areas: Mornington (10 min drive to venue) Mount Martha (15 min drive) Safety Beach (15 min drive) Dromana (15 min drive) Rosebud (20 min drive)
